Product Description

Plant Preservative Mixture (PPM™) is a broad-spectrum biocide developed specifically for plant tissue culture. It provides a solution to the persistent challenge of microbial contamination originating from airborne, waterborne and endogenous sources.

When used alongside appropriate aseptic laboratory techniques, PPM™ can help improve the success of plant tissue culture experiments by reducing microbial contamination and supporting the maintenance of healthy in vitro cultures.

Applications

Airborne Contamination Control Helps control microbial contamination introduced through the air.
Waterborne Contamination Management Supports the management of microorganisms introduced through water.
Endogenous Contamination Inhibition Helps reduce internal microbial contamination in plant explants.
In Vitro Seed Germination Helps maintain seed viability during tissue culture procedures.
Callus Proliferation Supports callus growth under appropriate culture conditions.
Callus Regeneration Supports regeneration processes in plant tissue culture.
Culture Media Stability Helps maintain the stability of tissue culture media.
Culture Experiment Success Helps improve the reliability of plant culture experiments.

Assay Protocol & Recommended Dosage

1. General Dosage Levels

With the exception of endogenous contamination, the recommended dosage range is 0.05%–0.2%. For callus proliferation, organogenesis and embryogenesis, the recommended range is 0.05%–0.075%. Higher concentrations may be needed to address higher levels of endogenous contamination.

2. Endogenous Contamination – Explants

Gently and routinely shake or stir explants approximately 1 cm long or shorter for 4–12 hours in a 4%–5% v/v PPM™ solution supplemented with 3× MS-strength basal salts, without Tween 20.

Without rinsing, transfer the explants into culture medium supplemented with 0.05%–0.1% PPM™ for herbaceous plants or 0.2% PPM™ for woody plants.

3. Endogenous Contamination – Tubers, Bulbs and Scales

Shake or stir the entire tuber, bulb or scale in bleach, then rinse with water. This rinsing step may be performed under non-sterile conditions. Slice the material into thin sections.

Shake or stir the slices for 12–24 hours in a 4%–5% PPM™ solution supplemented with full-strength basal salts, without pH adjustment and without Tween 20.

Without rinsing, transfer the material into culture medium supplemented with 0.1%–0.2% PPM™.

Protocol note: The tuber, bulb and scale procedure is indicated for ornamental plants in the supplied protocol. Follow the complete manufacturer's protocol and validate the dosage for the plant species and culture system.
